In-vitro Untersuchung zur sequenzspezifischen Therapie der Leichtketten-Amyloidose

In TP 6 werden die Aggregation von Leichtketten, die aus dem Blut und Urin von Studien-Patienten gewonnen wurden und die Wirksamkeit von EGCG auf diesen Prozess untersucht. Im nächsten Schritt sollen die Bindungstellen von EGCG an den Leichtketten identifiziert werden. Die genetischen und klinischen Ergebnisse der Studie (TP1) sollen mit der Aggregationskinetik und Toxizität in Beziehung gesetzt werden.


 

Sequence/specific intervention into light chain amyloidosis

Subproject 6 aims to correlate the amino acid sequence of light chain (LC) peptides with their aggregation propensity and the effect that treatment with epi-gallocatechine gallate (EGCG) has on their aggregation. Specifically, LC-peptides will be isolated from blood and urine of LC patients, and their aggregation kinetics will be recorded. EGCG will be tested for its ability to inhibit amyloid formation and remodel LC into aggregates that do not catalyze amyloid formation.  Finally, new potential therapeutic compounds from TP7 will be tested for their efficacy on different LC peptide sequences.
