Identifizierung neuer kleiner modulierender Moleküle für die Leichtketten-Aggregation bei AL Amyloidose

In TP 7 sollen zellfreie und zellbasierte Assays für die Identifikation von Modulatoren der Amyloidbildung etabliert werden. Ein systematisches Screening von chemischen Verbindungen, u.a. Derivate von EGCG, die in anderen Krankheitsmodellen die Aggregation beeinflusst haben, soll durchgeführt werden. Positiv getestete Substanzen sollen biochemisch, biophysikalisch und zellbiologisch charakterisiert werden, um eine Wirkung auf Zwischenprodukte des Aggregationsprozess nachzuweisen.

Identification of new small molecule modulators for light chain aggregation

Subproject 7 aims to establish cell-free and cell-based assays for the identification of chemical modulators of aggregation in light chain amyloidosis. A systematic screening of chemical compounds, partly derivatives of EGCG, will be carried out. Active molecules will be characterised and validated with biochemical, biophysical and cell biological methods. An important aspect of the investigations will be to detect effects on distinct intermediates of the aggregation cascade. In this way we hope to gather insight in the mechanisms of aggregation and the structure-activity relationship of modulator molecules.
